A significant minority (5-10%) of breast carcinomas is considered of

hereditary origin. Part of them is associated to mutations in the BRCA1
&lt;br /&gt;
and BRCA2 susceptibility genes, which have been extensively studied.

For their part, tumours in the non-BRCA1/BRCA2 breast cancer (BRCAx)
&lt;br /&gt;
families are increasingly believed to originate from multiple genetic
&lt;br /&gt;
events, as no &amp;quot;BRCA3&amp;quot; gene has been identified. The existence of
&lt;br /&gt;
specific &amp;quot;portraits&amp;quot; among hereditary breast carcinomas (HBC) has been
&lt;br /&gt;
questioned. Phenotype studies have shown that most BRCA1 tumours are
&lt;br /&gt;
part of a &amp;quot;basal epithelial-like&amp;quot; group of lesions. In contrast, BRCA2
&lt;br /&gt;
and BRCax HBC are more heterogenous, as also observed with sporadic
&lt;br /&gt;
carcinomas. Genetic analysis has also been applied to HBC, notably with
&lt;br /&gt;
the objective of resolving the heterogeneity of BRCAx lesions. This
&lt;br /&gt;
chapter aims to summarise recent data on BRCA1, BRCA2, and BRCAx HBC,

and to provide hypotheses on the origin of BRCA1 lesions, their

&lt;br /&gt;
who prescribes Tegretol and Depakote (both need blood
&lt;br /&gt;
levels monitored) he was concerned how/why my
&lt;br /&gt;
hemoglobin was low yet my Iron was high.
&lt;br /&gt;

&lt;br /&gt;
After finding a Hematologist &amp;amp; Gastro and having a
&lt;br /&gt;
slew of blood, genetic tests, a CAT scan &amp;amp; Liver Biopsy
&lt;br /&gt;
this is where I stand:
&lt;br /&gt;

&lt;br /&gt;
1. Beta Thalassemia trait (throughout family on mother's side);
&lt;br /&gt;
2. mild anemia;
&lt;br /&gt;
3. positive for presence of H63D mutation in the HFE gene, negative for
&lt;br /&gt;
C282Y mutation;
&lt;br /&gt;
4. Hemachromotosis / HHC Chromosome 6;
&lt;br /&gt;
5. too much Iron in my liver;
&lt;br /&gt;
6. being transitioned to a liver-safe epilepsy
&lt;br /&gt;
medication Keppra versus the Depakote.
&lt;br /&gt;

&lt;br /&gt;
My liver function tests (LFT's) improved dramatically after a
&lt;br /&gt;
phlebetomy and/or
&lt;br /&gt;
transitioning from the Depakote to the Keppra.....
&lt;br /&gt;

&lt;br /&gt;
I'm curious if there are any other cases of this combination which
&lt;br /&gt;
baffled my doctors for a few months until the genetic test, CAT and
&lt;br /&gt;
biospy.........
&lt;br /&gt;

&lt;br /&gt;
And what risks if any are there for having a family ??
&lt;br /&gt;

specimens suspected of infection with select agents in your lab?  By
&lt;br /&gt;
select agents, I mean anthrax, avian flu, tularemia, etc. as defined by
&lt;br /&gt;
the CDC.  I am currently in the process of drafting a policy for the
&lt;br /&gt;

Researchers have discovered that a drug used to treat tuberculosis

apparently cures patients of a Parkinson's-like illness suffered by
&lt;br /&gt;
thousands of mineworkers, welders and others exposed to high levels of
&lt;br /&gt;
the metal manganese.

Newswise - Researchers have discovered that a drug used to treat
&lt;br /&gt;
tuberculosis apparently cures patients of a Parkinson's-like illness
&lt;br /&gt;
suffered by thousands of mineworkers, welders and others exposed to
&lt;br /&gt;
high levels of the metal manganese.
&lt;br /&gt;

&lt;br /&gt;
Manganese is used to produce steel alloys and as a coating on welding
&lt;br /&gt;
rods, among other industrial applications. It replaced lead decades ago
&lt;br /&gt;
as a component in unleaded gasoline, increasing the risks of manganese
&lt;br /&gt;
intoxication for the general public, said one of the researchers, Wei
&lt;br /&gt;
Zheng, a professor and University Faculty Scholar in Purdue's School of
&lt;br /&gt;
Health Sciences.
&lt;br /&gt;

&lt;br /&gt;
When manganese builds up in toxic levels in the body, people suffer
&lt;br /&gt;
from &amp;quot;occupational manganese parkinsonism,&amp;quot; which causes symptoms
&lt;br /&gt;
similar to Parkinson's disease. Victims experience hand tremors, poor
&lt;br /&gt;
coordination, unsteady gait and a masklike inability to show facial
&lt;br /&gt;
expressions, Zheng said.
&lt;br /&gt;

&lt;br /&gt;
Manganese contained in the coating of welding rods is released in
&lt;br /&gt;
fumes. Welders involved in manufacturing vehicles, tanks and ships are
&lt;br /&gt;
especially prone to manganese intoxication because they work in close
&lt;br /&gt;
quarters, increasing their exposure to the metal, Zheng said.
&lt;br /&gt;

&lt;br /&gt;
&amp;quot;There are about 430,000 welders in the United States alone, and even
&lt;br /&gt;
more in China, so manganese intoxication likely affects many people,
&lt;br /&gt;
including workers involved in manganese mining and steel production,&amp;quot;
&lt;br /&gt;
he said. &amp;quot;In Beijing, we found a high percentage of welders have these
&lt;br /&gt;
symptoms.&amp;quot;
&lt;br /&gt;

&lt;br /&gt;
While the condition's symptoms are similar to those of Parkinson's
&lt;br /&gt;
disease, the standard treatments for Parkinson's disease, including the
&lt;br /&gt;
drug levodopa, are not effective for manganese intoxication. A chemical
&lt;br /&gt;
compound called EDTA has been used to help patients eliminate manganese
&lt;br /&gt;
in the urine. The drug's effectiveness, however, is limited because it
&lt;br /&gt;
is water-soluble, preventing it from readily passing through membranes
&lt;br /&gt;
in the &amp;quot;blood-brain barrier,&amp;quot; layers of cells surrounding blood vessels
&lt;br /&gt;
that block substances from traveling from the blood into brain tissue.
&lt;br /&gt;

&lt;br /&gt;
Ten researchers from institutions around the world - including Purdue -
&lt;br /&gt;
conducted a 17-year medical follow-up study on a manganese-poisoned
&lt;br /&gt;
worker and about 80 other patients. The researchers learned that an
&lt;br /&gt;
aspirinlike drug called sodium para-aminosalicylic acid, or PAS,
&lt;br /&gt;
dramatically reduces symptoms on a long-term basis.
&lt;br /&gt;

&lt;br /&gt;
&amp;quot;The amazing thing is that this drug reverses Parkinson-type symptoms
&lt;br /&gt;
of manganese intoxication,&amp;quot; Zheng said. &amp;quot;We see remarkable improvement
&lt;br /&gt;
after treatment with this drug even 17 years later.&amp;quot;
&lt;br /&gt;

&lt;br /&gt;
PAS has been used for decades to treat tuberculosis and apparently can
&lt;br /&gt;
cross the blood-brain barrier because it is fat-soluble, or lipophilic.
&lt;br /&gt;
That's because the drug contains a structure known as a benzene ring,
&lt;br /&gt;
which enables it to penetrate the membranes.
&lt;br /&gt;

&lt;br /&gt;
Findings will appear in the June issue of the Journal of Occupational
&lt;br /&gt;
Environmental Medicine. The paper was written by Yue-Ming Jiang, Xue-An
&lt;br /&gt;
Mo, Feng-Qi Du, Xue Fu and Xia-Yan Zhu, from Guangxi Medical University
&lt;br /&gt;
in China; Hong-Yu Gao and Feng-Ling Liao, from Wuzhou Center for
&lt;br /&gt;
Disease Prevention and Control in China; Jin-Lan Xie from the Wuzhou
&lt;br /&gt;
Worker's Hospital in China; Enrico Pira from the University of Turin in
&lt;br /&gt;
Italy; and Zheng.
&lt;br /&gt;

&lt;br /&gt;
The research has focused on China because that country is a major
&lt;br /&gt;
manganese ore producer and provides one-third of the world's supply of
&lt;br /&gt;
steel alloys.
&lt;br /&gt;

&lt;br /&gt;
The paper includes data from research involving a female Chinese
&lt;br /&gt;
mineworker who suffered debilitating symptoms, including lack of
&lt;br /&gt;
coordination, trouble walking and writing, and a masklike appearance
&lt;br /&gt;
caused by tense facial muscles. The woman's symptoms nearly disappeared
&lt;br /&gt;
after treatment with PAS in 1987, and she remained free of symptoms
&lt;br /&gt;
when re-examined during a follow-up study in 2004.
&lt;br /&gt;

&lt;br /&gt;
The researchers suggest several possible mechanisms that enable the
&lt;br /&gt;
drug to reverse symptoms of the illness. One is that the drug may
&lt;br /&gt;
contain &amp;quot;chelating arms&amp;quot; that grab manganese.
&lt;br /&gt;

&lt;br /&gt;
&amp;quot;However, we are not just looking at this drug as a chelating compound,
&lt;br /&gt;
but also as an anti-inflammatory, like aspirin,&amp;quot; Zheng said.
&lt;br /&gt;
&amp;quot;Historically, we have believed that neurodegeneration is permanent and
&lt;br /&gt;
cannot be reversed, but PAS appears to shed light on a reversal
&lt;br /&gt;
mechanism.
&lt;br /&gt;

&lt;br /&gt;
&amp;quot;It may possibly repair neurons. If this is true, this would be a major
&lt;br /&gt;
finding, but further research will be needed to study this possibility.
&lt;br /&gt;
We think the bigger picture is that the drug might also be used as a
&lt;br /&gt;
treatment for Parkinson's disease, but much more work is needed to
&lt;br /&gt;
confirm this theory.&amp;quot;

Although administration of androstenediol (a metabolite of

unknown whether this metabolite has any salutary effects in preventing

alterations in immune cell cytokine production following a combined insult

of T-H and sepsis. To examine this, male rats underwent laparotomy,

hemorrhagic shock (mean BP 40mmHg for 90min) and resuscitation or sham
&lt;br /&gt;
operation. Androstenediol (1mg/kg BW i.v.) or vehicle was administered at
&lt;br /&gt;
the end of resuscitation. Twenty hrs after T-H or sham operation, sepsis
&lt;br /&gt;
was induced by cecal ligation and puncture (CLP). Five hours thereafter,
&lt;br /&gt;
plasma cytokine levels and cytokine production of various immune cells were
&lt;br /&gt;
determined. In a separate set of experiments, survival was monitored for 10
&lt;br /&gt;
days after the induction of sepsis. Administration of androstenediol
&lt;br /&gt;
markedly decreased plasma IL-6 and TNF-alpha levels following T-H and CLP.
&lt;br /&gt;
Furthermore, it prevented the increased production of IL-6 and TNF-alpha by
&lt;br /&gt;
Kupffer cells and alveolar macrophages and attenuated the decrease in IL-6
&lt;br /&gt;
and TNF-alpha production by splenic macrophages; however, it had no
&lt;br /&gt;
significant effects on the depressed IL-6 and TNF-alpha production by PBMC
&lt;br /&gt;
following T-H and CLP. The depressed IL-2 and IFN-gamma production by
&lt;br /&gt;
splenocytes under those conditions was attenuated by the administration of
&lt;br /&gt;
androstenediol. Furthermore, survival rate following T-H and subsequent

sepsis was improved by androstenediol treatment. Since androstenediol

administration following T-H attenuated cytokine production and reduced

mortality in a double-hit model of T-H and sepsis, this agent appears to be

a novel and useful adjunct for maintaining the immune cell functions

nociception of the same intensity? I guess the answer is yes because

[such as an injury to the periosteum of the sternum] whereas C-fiber

It seems to me that the sensation induced by A-delta fiber stimulation

paralysis [no amount of stimulation (whether neural or direct electric

stimulation of the muscle fibers) could cause these muscles to contract

&lt;br /&gt;
2. While my breathing muscles are not paralyzed, my voluntary control

of them is totally lost [this means that my autonomic nervous system

has complete control over my respiration].
&lt;br /&gt;

3. The motor nerves supplying my voluntary muscles -- including speech

muscles but excluding breathing muscles -- are also relaxed into total

paralysis [these motor nerves are hyper-polarized] and unable to

The neurosteroid dehydroepiandrosterone (DHEA) at 1 nM protects
&lt;br /&gt;
NMDA-/GABAA-receptor negative neural crest-derived PC12 cells from
&lt;br /&gt;
apoptosis. We now report that membrane-impermeable DHEA-BSA conjugate
&lt;br /&gt;
replaces unconjugated DHEA in protecting serum-deprived PC12 cells from
&lt;br /&gt;
apoptosis (IC50=1.5 nM). Protection involves phosphorylation of the
&lt;br /&gt;
prosurvival factor Src and induction of the anti-apoptotic protein Bcl-2
&lt;br /&gt;
and is sensitive to pertussis toxin. Binding assays of [3H]DHEA on isolated
&lt;br /&gt;
PC12 cell membranes revealed saturation within 30 min and binding of DHEA
&lt;br /&gt;
with a Kd of 0.9 nM. A similar binding activity was detectable in isolated
&lt;br /&gt;
membranes from rat hippocampus and from normal human adrenal chromaffin
&lt;br /&gt;
cells. The presence of DHEA-specific membrane binding sites was confirmed
&lt;br /&gt;
by flow cytometry and confocal laser microscopy of DHEA-BSA-FITC stained
&lt;br /&gt;
cells. In contrast to estrogens and progestins, glucocorticoids and
&lt;br /&gt;
androgens displaced DHEA from its membrane binding sites but with a 10-fold
&lt;br /&gt;
lower affinity than DHEA (IC50=9.3 and 13.6 nM, respectively). These agents
&lt;br /&gt;
acted as pure antagonists, blocking the antiapoptotic effect of DHEA as
&lt;br /&gt;
well as the induction of Bcl-2 proteins and Src kinase activation. In
&lt;br /&gt;
conclusion, our findings suggest that neural crest-derived cells possess
&lt;br /&gt;
specific DHEA membrane binding sites coupled to G proteins. Binding to
&lt;br /&gt;
these sites confers neuroprotection.
